Motivated by the problem of colocalization analysis in fluorescence microscopic imaging, we study in this paper structured detection of correlated regions between two random processes observed on a common domain. We argue that although intuitive, direct use of the maximum log-likelihood statistic suffers from potential bias and substantially reduced power, and introduce a simple size-based normalization to overcome this problem. We show that scanning with the proposed size-corrected likelihood ratio statistics leads to optimal correlation detection over a large collection of structured correlation detection problems.

Multiple hypothesis testing is a fundamental problem in high-dimensional inference, with wide applications in many scientific fields. In genome-wide association studies, tens of thousands of tests are performed simultaneously to find if any single-nucleotide polymorphisms (SNPs) are associated with some traits and those tests are correlated. When test statistics are correlated, false discovery control becomes very challenging under arbitrary dependence. In this article, we propose a novel method—based on principal factor approximation—that successfully subtracts the common dependence and weakens significantly the correlation structure, to deal with an arbitrary dependence structure.We derive an approximate expression for false discovery proportion (FDP) in large-scale multiple testing when a common threshold is used and provide a consistent estimate of realized FDP. This result has important applications in controlling false discovery rate and FDP. Our estimate of realized FDP compares favorably with Efron’s approach, as demonstrated in the simulated examples. Our approach is further illustrated by some real data applications. We also propose a dependence-adjusted procedure that is more powerful than the fixed-threshold procedure. Discrepancy is a kind of important measure used in experimental design. Recently, a so-called discrete discrepancy has been applied to evaluate the uniformity of factorial designs. In this paper, we review some recent advances on application of the discrete discrepancy to several common experimental designs and summarize some important results.

Value at Risk (VaR) is a fundamental tool for managing market risks. It measures the worst loss to be expected of a portfolio over a given time horizon under normal market conditions at a given confidence level. Calculation of VaR frequently involves estimating the volatility of return processes and quantiles of standardized returns. In this paper, several semiparametric techniques are introduced to estimate the volatilities of the market prices of a portfolio. In addition, both parametric and nonparametric techniques are proposed to estimate the quantiles of standardized return processes. The newly proposed techniques also have the flexibility to adapt automatically to the changes in the dynamics of market prices over time. Their statistical efficiencies are studied both theoretically and empirically. The combination of newly proposed techniques for estimating volatility and standardized quantiles yields several new

Folded concave penalization methods have been shown to enjoy the strong oracle property for high-dimensional sparse estimation. However, a folded concave penalization problem usually has multiple local solutions and the oracle property is established only for one of the unknown local solutions. A challenging fundamental issue still remains that it is not clear whether the local optimum computed by a given optimization algorithm possesses those nice theoretical properties. To close this important theoretical gap in over a decade, we provide a unified theory to show explicitly how to obtain the oracle solution via the local linear approximation algorithm. For a folded concave penalized estimation problem, we show that as long as the problem is localizable and the oracle estimator is well behaved, we can obtain the oracle estimator by using the one-step local linear approximation. In addition, once the oracle estimator is obtained, the local linear approximation algorithm converges, namely, it produces the same estimator in the next iteration. The general theory is demonstrated by using four classical sparse estimation problems, that is, sparse linear regression, sparse logistic regression, sparse precision matrix estimation, and sparse quantile regression.
