The stochasticity of gene expression is manifested in the fluctuations of mRNA and protein copy numbers within a cell lineage over time. While data of this type can be obtained for many generations, most mathematical models are unsuitable to interpret such data since they assume non-growing cells. Here we develop a theoretical approach that quantitatively links the frequency content of lineage data to subcellular dynamics. We elucidate how the position, height, and width of the peaks in the power spectrum provide a distinctive fingerprint that encodes a wealth of information about mechanisms controlling transcription, translation, replication, degradation, bursting, promoter switching, cell cycle duration, cell division, gene dosage compensation, and cell size homeostasis. Predictions are confirmed by analysis of single-cell Escherichia coli data obtained using fluorescence microscopy. Furthermore, by matching the experimental and theoretical power spectra, we infer the temperature-dependent gene expression parameters, without the need of measurements relating fluorescence intensities to molecule numbers.

In this article, we survey recent developments in defining the quasi-local mass in general relativity. We discuss various approaches and the properties and applications of the different definitions. Among the expected properties, we focus on the rigidity property: for a surface in the Minkowski spacetime, one expects that the mass should vanish. We describe the Wang-Yau quasi-local mass whose definition is motivated by this rigidity property and by the Hamilton-Jacobi analysis of the Einstein-Hilbert action. In addition, we survey recent results on the minimizing property the Wang-Yau quasi-local mass.

In this note, we study submanifold geometry of the Atiyah--Hitchin manifold, a double cover of the 2-monopole moduli space, which plays an important role in various settings such as the supersymmetric background of string theory. When the manifold is naturally identified as the total space of a line bundle over S^2, the zero section is a distinguished minimal 2-sphere of considerable interest. In particular, there has been a conjecture about the uniqueness of this minimal 2-sphere among all closed minimal 2-surfaces. We show that this minimal 2-sphere satisfies the ``strong stability condition" proposed in our earlier work, and confirm the global uniqueness as a corollary.

Previous work has shown that in a two-dimensional periodic medium under focusing or defocusing cubic nonlinearities, gap solitons in the form of low-amplitude and slowly modulated single-Bloch-wave packets can bifurcate out from the edges of Bloch bands. In this paper, linear stability properties of these gap solitons near band edges are determined both analytically and numerically. Through asymptotic analysis, it is shown that these gap solitons are linearly unstable if the slope of their power curve at the band edge has the opposite sign of nonlinearity
here focusing nonlinearity is said to have a positive sign, and defocusing nonlinearity to have a negative sign. An equivalent condition for linear instability is that the power of the gap solitons near the band edge is lower than the limit power value on the band edge. Through numerical computations of the power curves, it is found that this condition is always satisfied, thus two-dimensional gap solitons near band edges are linearly unstable. The analytical formula for the unstable eigenvalue of gap solitons near band edges is also asymptotically derived. It is shown that this unstable eigenvalue is proportional to the cubic power of the soliton’s amplitude, and it induces width instabilities of gap solitons. A comparison between this analytical eigenvalue formula and numerically computed eigenvalues shows excellent agreement.

We propose a geometric inequality for two-dimensional spacelike surfaces in the Schwarzschild spacetime. This inequality implies the Penrose inequality for collapsing dust shells in general relativity, as proposed by Penrose and Gibbons. We prove that the inequality holds in several important cases.
