This article deals with statistical inferences based on the varying-coefficient models proposed by Hastie and Tibshirani. Local polynomial regression techniques are used to estimate coefficient functions, and the asymptotic normality of the resulting estimators is established. The standard error formulas for estimated coefficients are derived and are empirically tested. A goodness-of-fit test technique, based on a nonparametric maximum likelihood ratio type of test, is also proposed to detect whether certain coefficient functions in a varying-coefficient model are constant or whether any covariates are statistically significant in the model. The null distribution of the test is estimated by a conditional bootstrap method. Our estimation techniques involve solving hundreds of local likelihood equations. An effective bandwidth selection method for local linear regression is proposed in Fan and Gijbels [1995, J. Roy. Statist. Soc. Ser. B, 57, 371394]. The method is based on the idea of the pre-asymptotic substitution and has been tested extensively. This paper investigates the rate of convergence of this method. In particular, we show that the relative rate of convergence is of order <i>n</i>^2/7 if the locally cubic fitting is used in the pilot stage, and the rate of convergence is <i>n</i>^2/5 when the local polynomial of degree 5 is used in the pilot fitting. The study also reveals a marked difference between the bandwidth selection for nonparametric regression and that for density estimation: The plug-in approach for the latter case can admit the root-<i>n</i> rate of convergence while for the former case the best rate is of order <i>n</i>^2/5.

Motivated by recent work on studying massive imaging data in various neuroimaging studies, we propose a novel spatially varying coefficient model (SVCM) to capture the varying association between imaging measures in a three-dimensional volume (or two-dimensional surface) with a set of covariates. Two stylized features of neuorimaging data are the presence of multiple piecewise smooth regions with unknown edges and jumps and substantial spatial correlations. To specifically account for these two features, SVCM includes a measurement model with multiple varying coefficient functions, a jumping surface model for each varying coefficient function, and a functional principal component model. We develop a three-stage estimation procedure to simultaneously estimate the varying coefficient functions and the spatial correlations. The multiple testing procedure plays an important role in detecting the presence of spatial signals for large scale imaging data. Typically, the spatial signals are sparse but clustered. This paper provides empirical evidence that for a range of commonly used control levels, the conventional FDR procedure can lack the ability to detect statistical significance, even if the -values under the true null hypotheses are independent and uniformly distributed; more generally, ignoring the neighboring information of spatially structured data will tend to diminish the detection effectiveness of the FDR procedure. This paper first introduces a scalar quantity to characterize the extent to which the lack of identification phenomenon(LIP) of the FDR procedure occurs. Second, we propose a new multiple comparison procedure, called FDR, to accommodate the spatial information of neighboring -values, via a local aggregation of -values. Theoretical properties of the FDR procedure are investigated under weak dependence of -values. It is shown that the FDR procedure alleviates the LIP of the FDR procedure, thus substantially facilitating the selection of more stringent control levels. Simulation evaluations indicate that the FDR procedure improves the detection sensitivity of the FDR procedure with little loss in detection specificity. The computational simplicity and detection effectiveness of the FDR procedure are illustrated through a real brain fMRI dataset.

Local quasi-likelihood estimation is useful for nonparametric modeling in a widely-used exponential family of distributions, called generalized linear models. Yet, the technique cannot be directly applied to situations where a response variable is missing at random. Three local quasi-likelihood estimation techniques are introduced: the local quasi-likelihood estimator using only complete-data; the locally weighted quasi-likelihood method; the local quasi-likelihood estimator with imputed values. These estimators share basically the same first order asymptotic biases and variances. Our simulation results show that substantial efficiency gains can be obtained by using the local quasi-likelihood estimator with imputed values. We develop the local quasi-likelihood imputation methods for estimating the mean functional of the response variable. It is shown that the proposed mean imputation estimators are asymptotically normal with asymptotic variance that can be easily estimated. Data from an ongoing environmental epidemiologic study is used to illustrate the proposed methods.
