Measuring conditional dependence is an important topic in statistics with broad applications including graphical models. Under a factor model setting, a new conditional dependence measure based on projection is proposed. The corresponding conditional independence test is developed with the asymptotic null distribution unveiled where the number of factors could be high-dimensional. It is also shown that the new test has control over the asymptotic significance level and can be calculated efficiently. A generic method for building dependency graphs without Gaussian assumption using the new test is elaborated. Numerical results and real data analysis show the superiority of the new method.

Large-scale multiple testing with correlated and heavy-tailed data arises in a wide range of research areas from genomics, medical imaging to finance. Conventional methods for estimating the false discovery proportion (FDP) often ignore the effect of heavy-tailedness and the dependence structure among test statistics, and thus may lead to inefficient or even inconsistent estimation. Also, the commonly imposed joint normality assumption is arguably too stringent for many applications. To address these challenges, in this article we propose a factor-adjusted robust multiple testing (FarmTest) procedure for large-scale simultaneous inference with control of the FDP. We demonstrate that robust factor adjustments are extremely important in both controlling the FDP and improving the power. Discrepancy is a kind of important measure used in experimental design. Recently, a so-called discrete discrepancy has been applied to evaluate the uniformity of factorial designs. In this paper, we review some recent advances on application of the discrete discrepancy to several common experimental designs and summarize some important results.

This paper discusses statistical methods for estimating complex correlation structure from large pharmacogenomic datasets. We selectively review several prominent statistical methods for estimating large covariance matrix for understanding correlation structure, inverse covariance matrix for network modeling, large-scale simultaneous tests for selecting significantly differently expressed genes and proteins and genetic markers for complex diseases, and high dimensional variable selection for identifying important molecules for understanding molecule mechanisms in pharmacogenomics. Their applications to gene network estimation and biomarker selection are used to illustrate the methodological power. Several new challenges of Big data analysis, including complex data distribution, missing data, measurement error, spurious correlation, endogeneity, and the need for robust statistical methods, are also discussed.

We propose a bootstrap-based robust high-confidence level upper bound (Robust H-CLUB) for assessing the risks of large portfolios. The proposed approach exploits rank-based and quantile-based estimators, and can be viewed as a robust extension of the H-CLUB procedure (Fan etal., 2015). Such an extension allows us to handle possibly misspecified models and heavy-tailed data, which are stylized features in financial returns. Under mixing conditions, we analyze the proposed approach and demonstrate its advantage over H-CLUB. We further provide thorough numerical results to back up the developed theory, and also apply the proposed method to analyze a stock market dataset.
