In the analysis of cluster data the regression coefficients are frequently assumed to be the same across all clusters. This hampers the ability to study the varying impacts of factors on each cluster. In this paper, a semiparametric model is introduced to account for varying impacts of factors over clusters by using cluster-level covariates. It achieves the parsimony of parametrization and allows the explorations of nonlinear interactions. The random effect in the semiparametric model accounts also for within cluster correlation. Local linear based estimation procedure is proposed for estimating functional coefficients, residual variance, and within cluster correlation matrix. The asymptotic properties of the proposed estimators are established and the method for constructing simultaneous confidence bands are proposed and studied. Several new tests are proposed for examining the adequacy of a family of parametric models against large nonparametric alternatives. These tests formally check if the bias vector of residuals from parametric fits is negligible by using the adaptive Neyman test and other methods. The testing procedures formalize the traditional model diagnostic tools based on residual plots. We examine the rates of contiguous alternatives that can be detected consistently by the adaptive Neyman test. Applications of the procedures to the partially linear models are thoroughly discussed. Our simulation studies show that the new testing procedures are indeed powerful and omnibus. The power of the proposed tests is comparable to the <i>F</i>-test statistic even in the situations where the <i>F</i> test is known to be suitable and can be far more powerful than the <i>F</i>-test statistic in other situations. Improving efficiency for regression coefficients and predicting trajectories of individuals are two important aspects in the analysis of longitudinal data. Both involve estimation of the covariance function. Yet challenges arise in estimating the covariance function of longitudinal data collected at irregular time points. A class of semiparametric models for the covariance function by that imposes a parametric correlation structure while allowing a nonparametric variance function is proposed. A kernel estimator for estimating the nonparametric variance function is developed. In survival analysis, the relationship between a survival time and a covariate is conveniently modeled with the proportional hazards regression model. This model usually assumes that the covariate has a log-linear effect on the hazard function. In this paper we consider the proportional hazards regression model with a nonparametric risk effect. We discuss estimation of the risk function and its derivatives in two cases: when the baseline hazard function is parametrized and when it is not parametrized. In the case of a parametric baseline hazard function, inference is based on a local version of the likelihood function, while in the case of a nonparametric baseline hazard, we use a local version of the partial likelihood. This results in maximum local likelihood estimators and maximum local partial likelihood estimators, respectively. We establish the asymptotic normality of the estimators. An effective bandwidth selection method for local linear regression is proposed in Fan and Gijbels [1995, J. Roy. Statist. Soc. Ser. B, 57, 371394]. The method is based on the idea of the pre-asymptotic substitution and has been tested extensively. This paper investigates the rate of convergence of this method. In particular, we show that the relative rate of convergence is of order <i>n</i>^2/7 if the locally cubic fitting is used in the pilot stage, and the rate of convergence is <i>n</i>^2/5 when the local polynomial of degree 5 is used in the pilot fitting. The study also reveals a marked difference between the bandwidth selection for nonparametric regression and that for density estimation: The plug-in approach for the latter case can admit the root-<i>n</i> rate of convergence while for the former case the best rate is of order <i>n</i>^2/5.
